Papaverine

General characteristics. Structure:

Active ingredient: 0,02 g of a papaverine hydrochloride.

Excipients (basis): paraffin oil the P-1 type (or P-2), fat firm (витепсол W35) – before receiving suppository weighing 1,25 g.

Pharmacological properties:

Pharmacodynamics. Reduces a tone of unstriated muscles and renders in this regard vasodilating and spasmolytic action.

Is inhibitor of enzyme of phosphodiesterase and causes intracellular accumulation cyclic 3,5 adenosinemonophosphates and decrease in content of calcium that leads to disturbance of contractility of unstriated muscles and their relaxation. Action of a papaverine of a hydrochloride on the central nervous system is expressed poorly, only in high doses it renders sedation. In high doses reduces excitability of a cardiac muscle and slows down endocardiac conductivity.

Indications to use:

Spasm of peripheral vessels (endarteritis), brain vessels, stenocardia (as a part of complex therapy), a bronchospasm, a spasm of smooth muscles of abdominal organs (a pylorospasm, a spastic colitis, cholecystitis, cholelithiasis attacks), a spasm of uric ways, renal colic.

Route of administration and doses:

Rektalno, the adult for 0,02-0,04 (1-2 candles) 2–3 times a day.

Features of use:

During treatment alcohol intake has to be excluded. Vazodilatiruyushchy action decreases at tobacco smoking.

Use at pregnancy and a lactation. Use of drug during pregnancy and during breastfeeding safety of drug is not established

Side effects:

Allergic reactions, partial or full AV blockade, ventricular premature ventricular contraction, a lowering of arterial pressure (ABP), a lock, drowsiness, increase in activity of "hepatic" transaminases, an eosinophilia are possible.

Inform the attending physician on emergence in you of any undesirable or unpleasant reactions and feelings.

Interaction with other medicines:

Reduces protivoparkinsonichesky effect of a levodopa and hypotensive effect Methyldopums.

In a combination with barbiturates spasmolytic action of a papaverine of a hydrochloride amplifies.

At combined  use with tricyclic antidepressants, procaineamide, Reserpinum, quinidine sulfate strengthening of hypotensive effect is possible.

Contraindications:

Hypersensitivity to drug, atrioventricular (AV) blockade, glaucoma, a heavy liver failure, advanced age (risk of development of a hyperthermia), children's age till 1 year.

With care. A state after a craniocereberal injury, a chronic renal failure, insufficiency of function of adrenal glands, a hypothyroidism, a prostate hyperplasia, supraventricular tachycardia, depressed cases.

Overdose:

Symptoms: diplopia, weakness, decrease in the ABP, drowsiness.

Treatment: symptomatic (maintenance of the ABP).

Storage conditions:

In the dry, protected from light place at a temperature from 12-15 °C. In the place, unavailable to children. Period of validity 2 years. Not to use after the expiry date specified on packaging.

Suppositories rectal 0,02 g of 5 suppositories in a blister strip packaging. 2 planimetric packagings together with the application instruction place in a pack from a cardboard.
