Tiaprid

General characteristics. Structure:

Active agent: the tiaprida a hydrochloride of 111,10 mg corresponds to 100 mg of a tiaprid.

Pharmacological properties:

Tiaprid — an atypical neuroleptic, Sulpiridum analog. Tiaprid selectively blocks dopaminergic D2 receptors. Does not make impact on serotoninovy, adrenergic, histamine and muskarinovy receptors. Has weak hypotensive effect and weak extrapyramidal symptoms; does not render either sedative, nor anticholinergic influence. Anxiolytic action of a tiaprid was proved on several animal stressful models. Also it was proved what тиаприд has positive effect on a condition of wakefulness of patients in old age. The mechanism of its anxiolytic action was not explained yet, but it differs from its anti-dopaminergic activity.

Indications to use:

Adults: short-term therapy of behavior disorders in old age or at an alcoholism, the intensive, not leaving pain, dyskinesia, the senseless movements. Adults and children since 6 years: horea, Turett's syndrome. Children since 6 years: heavy behavior disorders, such as agitation and aggression.

Route of administration and doses:

Treatment should be begun with the minimum operating dose which then has to increase gradually. At short-term therapy of conditions of agitation and aggression, first of all in old age or at an alcoholism, the usual dose makes 200 — 300 mg/days within 1 — 2 months. Horea, Turett's syndrome: Adults: 300 — 800 mg/days. Children's age of 6 years: 3 — 6 mg/kg/. The intensive, not leaving pain at adults: 200 — 400 mg/days. Behavior disorders at children aged from 6 years, such as agitation and aggression: 100 — 150 mg/days. Tablets are washed down with small amount of liquid, they can be accepted with food or without it.

Features of use:

Tiaprid can cause an antipsychotic malignant syndrome, increase risk of developing of a stroke, venous thromboembolism or extend QT interval. Administration of drug during pregnancy is possible only in the most necessary cases as there is a risk of development of undesirable actions in newborns whose mothers accepted тиаприд in the third trimester of pregnancy. Reception of a tiaprid during feeding by a breast is not recommended. Ability of driving of motor transport and control of mechanisms can be temporarily reduced. Drug contains lactose.

Side effects:

Feeling of increased fatigue, drowsiness, sleeplessness, agitation, apathy, dizzinesses, headaches, extrapyramidal symptoms, endocrine disturbances owing to a giperprolaktinemiya (a galactorrhoea, a gynecomastia), concern, dystonia, increase in body weight.

Interaction with other medicines:

Contraindicated combinations: levodopa. Not recommended combinations: the alcohol, drugs containing the alcohol, drugs increasing risk of developing of tachycardia like "pirouette" (torsades de pointes) (some antiaritmik, drugs causing bradycardia or a gipokalemiya). The combinations demanding care: substances with overwhelming impact on TsNS, antagonists of H1 receptors, lithium.

Contraindications:

The known hypersensitivity to a tiaprid or to any of excipients of this drug, prolaktinzavisimy tumoral diseases, a pheochromocytoma, a concomitant use with a levodopa.

Overdose:

There are no data on overdose.

Storage conditions:

In the place protected from light, at a temperature not above 30 °C.
To store in the place, unavailable to children.
