General characteristics. Structure:

Pharmacological properties:

Pharmacodynamics. Myotropic spasmolysant, derivative isoquinoline. Inhibits phosphodiesterase IV that leads to accumulation of intracellular cyclic adenosinemonophosphate and as a result to an inactivation of an easy chain of a kinase of a myosin therefore there is a relaxation of smooth muscles.
Irrespective of type of a vegetative innervation, Drotaverinum affects unstriated muscles in digestive tract, bilious, urinogenital and vascular systems. In a myocardium and vessels the enzyme hydrolyzing cyclic adenosinemonophosphate is phosphodiesterase III that explains lack of serious side effects from cardiovascular system and not expressed therapeutic action concerning cardiovascular system.

Pharmacokinetics. It is quickly and fully soaked up in digestive tract. Bioavailability at intake is close to 100%, the period of semi-absorption of-12 min. It is evenly distributed on fabrics, gets into smooth muscle cells. It is generally removed by kidneys, to a lesser extent - with bile. Does not get through a blood-brain barrier.

Indications to use:

- the spasms of smooth muscles connected with diseases of a biliary path (a cholecystolithiasis, a cholangiolithiasis, cholecystitis, a pericholecystitis, a papillitis);
- spasms of smooth muscles of an uric path (nephrolithiasis, ureterolithiasis, pyelitis, cystitis, bladder tenesmus).
As auxiliary therapy:
- at spasms of smooth muscles of digestive tract (a peptic ulcer of a stomach and duodenum, gastritis, spasms of the cardia and the gatekeeper, enteritis, colitis, a spastic colitis with a lock and mucous colitis with a meteorism);
- tensor headache;
- at gynecologic diseases (dysmenorrhea).

Route of administration and doses:

Inside. The adult on 40-80 mg (1-2 tablets) 2-3 times a day.

Features of use:

At treatment of a peptic ulcer of a stomach and a 12-perstny gut apply in combination with other antiulcerous medicines.

Side effects:

Gastrointestinal disturbances (nausea, lock), disturbances of a nervous system (headache, dizziness, sleeplessness); cardiovascular disturbances (tachycardia, arterial hypotension), allergic reactions (including allergic dermatosis).

Interaction with other medicines:

Strengthens effect of other spasmolysants (including m-holinoblokatorov), the arterial hypotension caused by tricyclic antidepressants, quinidine, procaineamide. Reduces spazmogenny activity of morphine, protivoparkinsonichesky properties of a levodopa. Spasmolytic action of Drotaverinum increases at simultaneous use with phenobarbital.

Contraindications:

Hypersensitivity to active ingredient or any of drug components, a heavy renal or liver failure, heavy heart failure (a syndrome of low cordial emission), the lactation period, children's age up to 18 years. It is not necessary to accept to the people having a lactose intolerance, deficit of lactase or glyukozo-galaktozny malabsorption.
With care – at arterial hypotension, the expressed atherosclerosis of coronary arteries, a prostate hyperplasia, glaucoma, during pregnancy.

Overdose:

Atrioventricular block, cardiac standstill, paralysis of a respiratory center.
Treatment: drug withdrawal, symptomatic therapy. The specific antidote does not exist.

Storage conditions:

In dry, protected from light, the place, unavailable to children. Period of validity: 3 years. Not to use after the term specified on packaging.
