General characteristics. Structure:

Active agent: 1 tablet contains - a dezloratadina of 5 mg;

excipients: a mannitol, the hydroxypropyl cellulose low-replaced starch corn, talc, calcium hydrophosphate anhydrous, E 15 hydroxypropyl-methyl cellulose, titanium dioxide, propylene glycol, ferrous oxide red, ferrous oxide yellow.

Pharmacological properties:

Dezloratadin is primary active metabolite of a loratadin. Dezloratadin is the tricyclic antagonist of a histamine of long action, selectively blocks peripheral histamine _H1 – receptors and the cascade of cytotoxic reactions oppresses (release of cytokines _{_L4}, _{_L6}, _{_L8}, _{_L13}, chemokines like RANTES, products of superoxidic anion the activated polimorfoyaderny neutrophils, adhesion and a chemotaxis of eosinophils, IgE – dependent allocation of a histamine, D2 prostaglandin and C4 leukotriene; an acute allergic bronchospasm in researches on animals). Bioavailability is proportional to a dose (in the range of doses from 5 mg to 20 mg). Is defined in plasma in 30 minutes after reception. The maximum concentration in plasma after one-time reception in a dose of 5 or 7,5 mg is reached in 3 h. Contacts proteins of plasma of 83 - 87%. The elimination half-life _(T1/2) averages 27 hours. Does not get through a blood-brain barrier, passes through a placental barrier and gets to breast milk. It is metabolized in an organism by a hydroxylation and a glyukuronization, removed with urine (less than 2% - in steady-state condition) and with excrements (less than 7% - in steady-state condition). Is not removed at a hemodialysis. At reception in dosing of 5 - 20 mg within a week cumulation is not observed.

Indications to use:

Seasonal allergic rhinitis and pollinosis. Chronic idiopathic small tortoiseshell.

Route of administration and doses:

Apply inside. Accept irrespective of meal, without chewing, washing down with water. Adults and children are more senior than 12 years accept on 1 tablet of times a day. Adults with a renal or liver failure are recommended to begin treatment with a dose of 5 mg; adjusting the subsequent doses of reception depending on clinical performance. The drug dosing for children with a renal or liver failure was not studied. It is regularly desirable to accept drug, at the same time days. Duration of treatment is defined by weight and the course of a disease.

Features of use:

Use during pregnancy or feeding by a breast. Safety of use of drug of Alergo – Norms at pregnant women is not established therefore it is not recommended to appoint it during pregnancy. Dezloratadin gets into breast milk therefore women should not appoint drug during feeding a breast. 

Ability to influence speed of response at control of motor transport or other mechanisms. It was not noted.

Side effects:

Seldom – a headache (2%), fatigue (about 1%), dryness in a mouth (about 0,1%). Very seldom – hypersensitivity reactions (including an anaphylaxis and rashes), tachycardia, a heart consciousness, a psychomotor hyperactivity, spasms, increase in activity of liver enzymes, increase in level of bilirubin, development of hepatitis. In case of any undesirable phenomena the patient needs to consult to the doctor.

Contraindications:

Hypersensitivity to a dezloratadin or a loratadin, pregnancy, the feeding period a breast.

Overdose:

Symptoms: manifestation strengthening collateral effects. Treatment: symptomatic therapy; the hemodialysis is inefficient.
