Potassium orotate

General characteristics. Structure:

Chemical name: potassium salt of uracil-4-carboxylic (orotovy) acid;
main physical and chemical properties: tablets of white  color; structure: one tablet contains potassium of an orotat 0,5 g; excipients: sugar milk, potato starch, edible gelatin, stearic acid.

Pharmacological properties:

Pharmacodynamics. Anabolic means. Makes the general stimulating impact on exchange processes. Participates in proteinaceous, carbohydrate, lipidic exchanges.
Orotovy acid – one of predecessors of the pirimidinovy bases (uracil, thymine, цитозин) in the course of protein synthesis. Participation of orotovy acid in carbohydrate metabolism consists in its influence on exchange of a galactose. As connection of anabolic type of action, orotovy acid and its salts are applied to recovery of disturbances of protein metabolism and as a stimulator of processes of metabolism.
Drug promotes strengthening of a diuresis.

Pharmacokinetics. Drug is absorbed in a digestive tract not completely, about 10% of the accepted dose are soaked up. In a liver turns into orotidin-5-phosphate. More than 30%, doses that is soaked up, removed with urine in the form of various metabolites.

Indications to use:

Potassium оротат apply in complex therapy in combination with other medicines (vitamins, cardiotonic drugs, etc.); at the diseases of a liver caused by acute and chronic intoxications (except for liver cirrhoses with ascites); at dystrophy and a myocardial infarction, nutritional and alimentary and infectious dystrophy at children, at anemias, in the period of reconvalescence, at chronic heart failure, at arrhythmias (especially ciliary arrhythmia and premature ventricular contraction) and other states demanding stimulation of anabolic processes including at the raised exercise stresses.

Route of administration and doses:

Potassium оротат appoint inside in 1 hour prior to food or in 4 hours after food. The adult appoint 0,5-1,5 g (1 – 3 tablets) a day (the daily dose is accepted in 2-3 receptions on 0,25-0,5 g). In some cases, depending on efficiency of drug, the daily dose for adults is increased to 3 g.
To children at nutritional and alimentary and infectious dystrophy, anemias, in the period of reconvalescence appoint Potassium оротат in a daily dose 10-20 mg (0,01-0,02) to body weight kg. The specified dose is accepted in 2-3 receptions.
Usually the course of treatment is equal to 3-5 weeks. If necessary the course of treatment is repeated after a monthly break.

Features of use:

For use in cardiological practice the combination of orotovy acid and magnesium is perspective.
Potassium оротат cannot be used as kaliysoderzhashchy drug, for kaliyzameshchayushchy therapy.

Side effects:

Tablets of Potassium of an orotat are usually well transferred. The allergic phenomena, dyspepsia which quickly pass at drug withdrawal are in some cases observed. In need of cases of emergence of by-effects appoint antihistamines.

Interaction with other medicines:

Efficiency of Potassium of an orotat increases at joint administration of folic acid and cyanocobalamine. Potassium оротат improves portability of cardiac glycosides.

Contraindications:

Hypersensitivity to drug, liver cirrhoses with ascites.

Overdose:

At use in high doses against the background of a low-proteinaceous diet development of dystrophy of a liver is possible that demands drug withdrawal.

Storage conditions:
