Mebeverin

General characteristics. Structure:

Active ingredient: 135 mg of a mebeverin in 1 tablet.

The drug having myotropic, spasmolytic effect.

Pharmacological properties:

Pharmacodynamics. Blocks bystry natrium channels of a cellular membrane of a myocyte and breaks an entrance of ions of Na + in a cell, blocks an entrance of ions of Ca2 + via slow channels, slows down processes of depolarization of a membrane and interferes with reduction of muscle fibers.

Directly relaxes smooth muscles, preferential a GIT. Eliminates a spasm, does not exert significant impact on a normal peristaltics of intestines. Has no anticholinergic effect.

Pharmacokinetics. After intake it is not found in plasma since is exposed to presistemny hydrolysis in a liver with formation of 3,4-dimetoksibenzoyny acid (veratric acid) and mebeverinovy alcohol. It is removed preferential with urine only in the form of metabolites — the veratric acid and mebeverinovy alcohol (including partially in the form of the corresponding carboxylic acid, partially — in the form of demetilirovanny carboxylic acid), in small amounts is defined in bile. It is completely removed during 24 h after reception of a single dose. Does not kumulirut.

Data of preclinical tests for animals. Researches of acute and subchronic toxicity, and also influence on reproductive function at different types of laboratory animals showed that LD50 at introduction of a mebeverin inside made 902–1980 mg/kg, at in introduction — 1% of LD50 at peroral introduction. The main symptoms observed after introduction by an animal of a mebeverin in high doses inside and parenterally demonstrated defeat of TsNS which sign was the excited behavior.

Teratogenic action is not revealed.

Indications to use:

At adults: a spasm of bodies of a GIT (including caused by an organic disease), intestinal colic, bilious colic; syndrome of the angry intestines (symptomatic treatment of pain and spasms, and also intestinal frustration and discomfort).

At children 12 years are more senior: the functional disorders of a GIT which are followed by an abdominal pain.

Route of administration and doses:

Inside, without chewing, in 20 min. prior to food, washing down with water, to adults and children 12 years are more senior appoint 200 mg 2 times a day.

Features of use:

Use at pregnancy and feeding by a breast. At pregnancy it is possible if the expected effect of therapy exceeds potential risk for a fruit. At use in therapeutic doses мебеверин does not get into breast milk.

Due to a possibility of development of dizziness during treatment it is necessary to observe extra care during the driving of vehicles, control of cars and mechanisms.

Side effects:

Dizziness (in some cases), diarrhea or lock; allergic reactions (small tortoiseshell, Quincke's edema, face edema and dieback).

Contraindications:

Hypersensitivity.

Overdose:

Symptoms: excitement of TsNS.

Treatment: removal of not absorbed drug from a GIT (a gastric lavage, purpose of absorbent carbon), a symptomatic and maintenance therapy. The specific antidote is unknown.

Storage conditions:

To store at a temperature of 15-25 °C, in dry, protected from light and the place, unavailable to children. Period of validity 3 years. Not to use after the expiry date specified on packaging.
