Kraynon

Kraynon – the gestagenny drug causing the transition of a mucous membrane of a uterus to a secretory phase from a phase of the proliferation caused by effect of follicle-stimulating hormone (FSG) reducing excitability and contractility of uterine tubes and muscles of a uterus.

Form of release and structure

Dosage form – gel vaginal: a soft consistence, homogeneous, white or almost white color, possesses a specific smell (on 1 dose in one-time polyethylene vaginal applicators, on 6 or 15 applicators in a pack cardboard).

Active agent: progesterone, contains in 1 applicator (1,125 g) – 90 mg.

Additional substances: the water purified glyceride of the hydrogenated palm-oil, paraffin easy liquid, carbomer 974P, polikarbofit, sorbic acid, sodium hydroxide, глицерол.

Indications to use

• Replaceable hormonal therapy in the period of a postmenopause (in a combination with estrogen);
• Dysfunctional uterine bleedings and secondary amenorrhea owing to deficit of progesterone.

Also Kraynon apply to maintenance of a lyuteinovy phase at use of auxiliary methods of a reproduction.

Contraindications

Absolute:

• Malignant tumors of generative organs or mammary glands or suspicion on their existence;
• Vaginal bleedings of not clear etiology;
• Incomplete abortion;
• Acute porphyria;
• Acute disorder of cerebral circulation now or in the anamnesis;
• Thromboembolic diseases, acute thrombophlebitises and fibrinferments;
• Lactation;
• Hypersensitivity to drug components.

Relative (at the following diseases / states Kraynon it is necessary to apply with extra care because of risk of development of complications):

• Arterial hypertension;
• Cardiovascular insufficiency;
• Chronic renal failure;
• Bronchial asthma;
• Migraine;
• Epilepsy;
• Giperlipoproteinemiya;
• Depression;
• Diabetes mellitus.

Route of administration and dosage

Kraynon is applied intravaginalno.

The recommended dosing modes:

• Replaceable hormonal therapy in a postmenopause: on 1 applicator (90 mg of progesterone) 2 times a week;
• Dysfunctional uterine bleedings and secondary amenorrhea owing to deficit of progesterone: on 1 applicator every other day from the 15th to the 25th day of a cycle. If necessary the dose is increased or reduced;
• Maintenance of a lyuteinovy phase at use of auxiliary methods of a reproduction: on 1 applicator daily from the date of transfer of an embryo within 10-12 weeks from the moment of confirmation of pregnancy or to the term of 12 weeks.

Rules of use of drug:

1. Having densely clamped the applicator in an upper part of a palm, between big and index fingers, to stir up it it is similar to the medical thermometer that gel moved to the lower end of the applicator;
2. Holding the applicator for the upper flat end of an air container, by turn, carefully, without pressing on a container, to remove the breaking-off lid since the opposite end;
3. To reach a comfortable position (lying with the legs which are slightly bent in knees or sitting) and to accurately enter the lower end of the applicator deeply into a vagina;
4. It is strong to squeeze an air container that drug got into a vagina. A quantity of gel will remain in the applicator, nevertheless the dose will be entered completely;
5. To throw out the used applicator with the remained gel.

Side effects

• From a reproductive system: intermenstrual bleedings, morbidity of mammary glands, irritation of a mucous membrane of a vagina and other reactions of moderate expressiveness in the field of application;
• From the central nervous system: drowsiness and headache;
• From the alimentary system: abdominal pain;
• Allergic reactions: hypersensitivity reactions (usually – skin rash).

Special instructions

Before appointment Kraynona the patient has to undergo the inspection at the doctor including the analysis of a cervical smear (Papanikolau's test), assessment of development and a condition of pelvic bodies and mammary glands.

Drug can be used in the I trimester of pregnancy at insufficiency of function of a yellow body. On later terms prescription of medicine is not recommended.

Patients with any risk factors of development of trombotichesky disturbances (an embolism of a pulmonary artery, disturbances of cerebral circulation, thrombophlebitis, eye retina thrombosis) have to be under careful observation.

During treatment it is necessary to pay special attention to possible precursory symptoms of trombotichesky disturbances. In case of any symptom allowing to suspect existence of such disturbance it is necessary to cancel immediately Kraynon.

At prolonged treatment regular gynecologic surveys are shown to exclude probability of development of a hyperplasia of an endometria.

In order to avoid risk of incomplete abortion at use of drug it is regularly necessary to conduct ultrasonography or to determine the level of a chorionic gonadotrophin.

In cases of irregular vulval bleeding, including break bleedings, it is necessary to exclude organic pathology.

At vaginal bleeding of not clear genesis it is necessary to conduct the corresponding examination.

Special observation during treatment is required to patients with a depression in the anamnesis. At emergence of depressive symptoms Kraynon's use should be interrupted.

In certain cases etrogen-progestagenny drugs can reduce tolerance to glucose therefore patients with a diabetes mellitus have to be under careful medical observation.

The sorbic acid which is Kraynon's part can cause contact dermatitis.

Drug can cause drowsiness and a feeling of fatigue therefore it is necessary to be careful when driving and performing potentially dangerous types of works (alcohol can strengthen this effect).

Medicinal interaction

Kraynon along with other intravaginalny means is not recommended to apply.

Terms and storage conditions

To store in the place, unavailable to children. Not to exceed temperature of 25 °C. Not to freeze.

Period of validity – 3 years.
