Aveksim's sulfaguanidine

General characteristics. Structure:

Active ingredient: 500 mg of Sulfaguanidinum monohydrate (sulfaguanidine).

Excipients: potato starch, calcium stearate.

Antimicrobic bacteriostatic means.

Pharmacological properties:

Pharmacodynamics. The mechanism of action is caused by antagonism with paraaminobenzoic acid (PABK) and competitive oppression of a digidropteroatsintetaza that leads to disturbance of synthesis of the tetrahydrofolic acid necessary for synthesis of purines and pyrimidines. Growth of colibacillus oppresses and reduces synthesis of thiamin, Riboflavinum, niacin in intestines. SULFAGUANIDINE has bacteriostatic action on Streptococcus spp., Staphylococcus spp., Escherichia coli, Shigella dysenteriae, Proteus vulgaris.

Pharmacokinetics. After intake drug is slightly absorbed from a digestive tract therefore there is a high bacteriostatic concentration in intestines.

Indications to use:

— bacterial dysentery;

— colitis and coloenteritis;

— a preparation for surgery on a large intestine.

Route of administration and doses:

At acute dysentery conduct 2 courses of treatment. First course: in the 1 and 2 day - 1 g each 4 h (only 6 g/days), 3 and 4 day - 1 g each 6 h (4 g/days), 5 and 6 day - 1 g each 8 h (3 g/days). If necessary in 5-6 days conduct the second year: 1 and 2 day - on 1 g each 4 h, at night - in 8 h (only 5 g/days), 3 and 4 day - 1 g in 4 h (4 g/days), do not give at night, 5 day - 3 g/days.

To children 3 years - 0.5 g 4 times/days within 7 days are more senior.

At treatment of other infections: the adult - inside, on 1-2 g each 4-6 h; in the next 2-3 days - half doses.

To children 3 years - in the first day on 0.1 g/kg/days equal doses each 4 h, in the next days - on 0.25-0.5 g each 6-8 h are more senior.

For prevention of postoperative complications from intestines - on 0:05 g/kg each 8 h, within 5 days to and 7 days after operation.

The highest doses for adults: one-time - 2 g, daily - 7 g.

Features of use:

Use of drug for children aged up to 3 years is forbidden.

At acute dysentery to children 3 years - 0.5 g 4 times/days within 7 days are more senior.

At treatment of other infections: to children 3 years - in the first day on 0.1 g/kg/days equal doses each 4 h, in the next days - on 0.25-0.5 g each 6-8 h are more senior.

For the purpose of the prevention of a crystalluria during treatment patients have to receive plentiful drink (2-3 l/days).

The early termination of treatment or insufficient doses can lead to emergence of resistant strains of activators.

During treatment reception of vitamins of group B is recommended.

Side effects:

From the alimentary system: nausea, vomiting.

From an urinary system: crystalluria.

Allergic reactions: urticaria, exfoliative dermatitis.

Others: a hypovitaminosis In, a leukopenia, neuritis.

Interaction with other medicines:

Reduces reliability of peroral contraception.

Benzocaine, Procainum reduce antimicrobic activity (at hydrolysis release PABK).

Non-steroidal anti-inflammatory drugs, hypoglycemic medicines (sulphonylurea derivatives), Phenytoinum and anticoagulants of indirect action increase expressiveness of side effects of drug.

Ascorbic acid, hexamethylenetetramine increase risk of development of a crystalluria.

Myelotoxic medicines strengthen manifestations of a gematotoksichnost of drug.

Contraindications:

— hypersensitivity to components of drug or other streptocides;

— deficit glyukozo-6-fosfatdegidrogenazy;

— нефроуролитиаз;

— porphyria;

— B12-scarce anemia;

— pregnancy;

— lactation period;

— children's age up to 3 years;

— a hyperbilirubinemia (at children).

Overdose:

Data on overdose are absent.

Storage conditions:

Drug should be stored in the place, dry, unavailable to children, at a temperature not above 30 °C. Period of validity 4 years. At expiry date not to use drug.
