General characteristics. Structure:

Active ingredient: tetrizolina hydrochloride.
Tizin of 0,1% for adults contains in 1 ml of drug 1,0 mg of a tetrizolin of a hydrochloride.
Tizin of 0,05% for children contains in 1 ml of drug 0,5 mg of a tetrizolin of a hydrochloride.

Excipients: sorbitol solution of 70% (uncrystallizable), a gipromelloza, benzyl alcohol, sodium citrate, oil perfumery (No. 25768), a chloride benzalkoniya solution of 17%, dinatrium эдетат, Acidum hydrochloricum (Cremophor 40) concentrated 37%, a macrogoal глицерилгидроксистеарат, water cleared.

Pharmacological properties:

Pharmacodynamics. Tetrizolin treats group of sympathomimetics and exerts the direct stimulating impact on alpha and adrenergic receptors of a sympathetic nervous system. At the same time it does not render or almost does not exert impact on beta and adrenergic receptors. As sympathomimetic amine drug has vasoconstrictive and antiedematous effect at hit on mucous a nose that leads to narrowing of small arterioles of the nasal courses, reduction of puffiness mucous a nose and to decrease in secretion.
Vasoconstrictive action and reduction of hypostasis mucous under the influence of a tetrizolin begins about 1 minute later after topical administration of drug and 4–8 hours proceed.

Pharmacokinetics. At topical administration and observance of recommendations about use of drug of absorption of a tetrizolin practically does not occur.

Indications to use:

For reduction of hypostasis mucous a nose at the patients having rhinitis, pharyngitis, sinusitis, hay fever and also for reduction of hypostasis mucous a nose in need of holding diagnostic and medical actions.

Route of administration and doses:

Tizin of a drop in a nose of 0,1% for adults
If there are no other instructions, then to adults and children 6 years are more senior appoint 2–4 drops of drug in each nostril of 3-5 times a day.
Tizin of a drop in a nose of 0,05% for children
If there are no other instructions, to children aged from 2 up to 6 years appoint 2–3 drops of drug in each nostril 3–4 times a day.

Tizin it is necessary to apply not more often than 1 time at 4 o'clock.
As a rule, more rare use of drug happens sufficient as in most cases drug has long effect (frequent till 8 o'clock). Single use of drug before going to bed removes a nose congestion on all night long, but can cause sleeplessness owing to impact of a tetrizolin on TsNS.
Tizin it is possible to apply within a short period of time. Duration of treatment has to make no more than 5 days for adults and no more than 3 days for children. Repeatedly it is possible to appoint drug only in several days.

Features of use:

Influence on ability to drive the car and to work with mechanisms

At too prolonged use of drug or use of the doses exceeding recommended development of system cardiovascular effects is possible. System side effects can lead to disturbance of ability to driving or work with mechanisms.

Side effects:

Reactive hyperemia, burning sensation mucous, chronic puffiness mucous a nose at prolonged use of drug, sometimes perhaps systemic action (heartbeat, headaches, sleeplessness, a tremor, weakness, perspiration, increase in arterial pressure, nausea, allergic reactions).
Too frequent use or long (more than 5-7 days) can lead use of a tetrizolin, and also use of drug in high doses to emergence of burning sensation and dryness mucous a nose, and also to development of reactive hypostasis and medicamentous rhinitis.
Prolonged use of derivatives of an imidazole can have the damaging effect on mucous, suppressing activity of a ciliate epithelium. Irreversible damage of mucous can lead to development of dry rhinitis.

Interaction with other medicines:

Combined use with inhibitors of a monoaminooxidase and tricyclic antidepressants can lead to increase in arterial pressure as a result of potentiation of vasoconstrictive effect.

Contraindications:

- Hypersensitivity to any of drug components;
- dry rhinitis;
- closed-angle glaucoma;
- Tizin of 0,1% for adults is contraindicated to children up to 6 years;
- Tizin of 0,05% for children is contraindicated to children up to 2 years.

With care: at patients with a serious cardiovascular illness (for example the coronary heart disease (CHD), arterial hypertension, arrhythmia, aneurism, a pheochromocytoma), with metabolism disturbances (for example a hyperthyroidism, diabetes) and at the patients accepting the MAO inhibitors or other drugs increasing arterial pressure, Tizin it is possible to apply only after careful comparison of possible advantage and risk to the patient.

Pregnancy and lactation

As there is a probability of emergence of system side effects (generally vasoconstrictive effect), use of drug during pregnancy and in the period of a lactation perhaps only after careful comparison of possible advantage for mother and risk for a fruit (child).

Overdose:

The risk of development of system side effects is high at small children at drug hit inside.
Excess system absorption of alpha and sympathomimetic derivatives of an imidazole can lead to oppression of the central nervous system that is shown by drowsiness, decrease in body temperature, bradycardia, hypotension, an apnoea and a coma.
The mydriasis, nausea, cyanosis, fever, spasms, tachycardia, cardiac arrhythmia, cardiac standstill, hypertensia, fluid lungs, respiratory insufficiency, mental disturbances belong to usual symptoms of overdose.
The antidote of a tetrizolin of a hydrochloride is unknown.
Treatment: purpose of absorbent carbon, gastric lavage, oxygen, and also febrifuges and anticonvulsant drugs in case of need. For the purpose of a lowering of arterial pressure appoint 5 mg of phentolamine on normal saline solution in/in slowly or 100 mg inside. Vasoconstrictive drugs are contraindicated at hypotension.
