General characteristics. Structure:

Active agent: 50 mg, 200 mg, 400 mg of an amisulprid.

Pharmacological properties:

Amisulprid — an atypical anti-psychotic with benzamide structure, Sulpiridum analog, with strong antagonistic effect on dopamine receptors of D2 and D3. Possesses two-phase action — in low doses blocks presynaptic dopamine receptors (it causes the dopamine release which is responsible for desingibitorny effect), and in higher doses blocks postsynaptic receptors. Does not make impact on serotoninovy, adrenergic, histamine and muskarinovy receptors. Has weak sedative and hypotensive effect and weak extrapyramidal symptoms; does not make anticholinergic impact.

Indications to use:

Drug is intended for treatment of acute and chronic schizophrenia. It is effective at treatment of positive symptoms of schizophrenia with nonsense, hallucinations, disturbances of thinking. Primary negative symptoms with inadequate affectivity, emotional manifestations, and also the loss of the patient from society accompanying schizophrenia.

Route of administration and doses:

At positive symptoms of 400 — 800 mg/days, in some cases 1 200 mg/days. At negative symptoms of 50 — 300 mg/days. At a dosage to 300 mg амисульприд can be accepted once a day, higher amount of drug has to be divided into two receptions.

Tablets / tablets, coated, accept regardless of meal.

Features of use:

Amisulprid can cause a malignant antipsychotic syndrome, increase risk of developing of a stroke, extend an interval of QT and worsen course of a disease of Parkinson. Amisulprid can strengthen impact of alcohol on TsNS therefore during treatment it is impossible to take alcohol. It is not recommended to use drug during pregnancy. The newborns subjected during the 3rd trimester to influence of an amisulprid have a risk of development of ekrstrapiramidny signs or signs of cancellation. At women decrease in fertility was observed. Ability of driving of motor transport and control of mechanisms can be temporarily reduced.

Side effects:

Usual reactions: sleeplessness, depression, agitation. More rare reactions: drowsiness, a lock, nausea, vomiting, dryness in a mouth. As well as at other anti-psychotics: increase in plasmatic levels of prolactin, increase in body weight, acute dystonia, extrapyramidal symptoms, tardivny dyskinesia, hypotension and bradycardia. In exceptional cases: lengthening of an interval of QT, tachycardia like "pirouette" (torsade de pointes), allergic reactions, increase in level of enzymes of a liver, epilepsy attacks.

Interaction with other medicines:

Contraindicated combinations: antiaritmik of group I and III, bepridit, цизаприд, сультоприд, thioridazine, erythromycin intravenously, винкамицин intravenously, галофантрин, pentamidine, спарфлоксацин, a levodopa, methadone. Not recommended combinations: the drugs increasing risk of development of heavy ventricular arrhythmias – the drugs causing bradycardia, neuroleptics, tricyclic antidepressants, lithium, астемизол, терфенадин, the drugs causing a gipokalemiya. The combinations demanding care: substances with sedative impact on TsNS, anti-hypertensive drugs.

Contraindications:

The known hypersensitivity to an amisulprid or any of excipients of this drug, prolaktinzavisimy tumoral diseases, a pheochromocytoma, patients with severe forms of a disease of kidneys, children up to 18 years, feeding by a breast. A concomitant use with drugs which can cause heavy ventricular arrhythmias, a combination to a levodopa.

Overdose:

There are no messages on overdose.
