Clotrimazolum

General characteristics. Structure:

Active   agent:   Clotrimazolum of 100 mg

Excipients: lactose (sugar milk) - 350,0 mg, starch corn - 255,0 mg, cellulose microcrystallic - 205,0 mg, Natrium benzoicum - 10,0 mg, gelatin - 40,0 mg, magnesium stearate - 10,0 mg, silicon dioxide colloid - 30,0 mg.

Pharmacological properties:

Clotrimazolum is antifungal broad-spectrum agent for topical administration. The anti-mycotic effect of Clotrimazolum (derivative an imidazole) is connected with disturbance of synthesis of the ergosterol which is a part of a cellular membrane of mushrooms that changes permeability of a membrane and causes the subsequent lysis of a cell. In small concentration affects fungistaticly, and in big — fungitsidno, and not only proliferating cells. In fungicidal concentration interacts with mitochondrial and peroksidazny enzymes therefore there is an increase in concentration of peroxide of hydrogen up to toxic level that also promotes destruction of fungal cells.
It is effective concerning dermatophytes (Trichophyton rubrum, Trichophyton mentagrophytes, Epidermophyton floc-cosum, Microsporum canis), barmy and mold mushrooms (the sort Candida, Torulopsis glabrata, the sort Rhodotorula), an also of the activator multi-colored depriving of Pityriasis versicolor (Malassezia furfur).

Pharmacokinetics. At use Clotrimazolum and intravaginalno absorption makes 3-10% of the entered dose. High concentration in a vaginal secret and low concentration in blood remain within 48-72 hours. In a liver it is quickly metabolized to inactive metabolites.

Indications to use:

— the genital infections caused by drozhzhepodobny mushrooms of the sort Candida  (a candidosis  vulvovaginitis);

Route of administration and doses:

Locally.
Only for intravaginalny use. To adults and teenagers (after the beginning of sex life) vaginal tablets enter in the evening into a vagina as it is possible more deeply, in a dorsal decubitus at slightly bent legs, daily within 6 days on 1 vaginal tablet on 100 mg. The repeated course of treatment is possible after consultation of the doctor.

Features of use:

Prevention of urogenital reinfection requires simultaneous treatment of sexual partners. At a simultaneous infection of vulvar lips and adjacent sites (a candidosis vulvitis) it is necessary to carry out in addition topical treatment by the drug Clotrimazolum cream.
In II, III trimesters of pregnancy treatment by vaginal tablets should be carried out without applicator. At patients with a liver failure it is necessary to control a functional condition of a liver periodically. At emergence of signs of hypersensitivity or irritation treatment is stopped.
In the absence of effect within 3 days it is necessary to confirm the diagnosis.

Side effects:

Itch, burning and hypostasis of a mucous membrane of a vagina, allocation from a vagina, a headache, a gastralgia, the speeded-up urination, intercurrent cystitis, a burning sensation in a penis at the sexual partner, pain during sexual intercourse.

Interaction with other medicines:

At simultaneous use with Amphotericinum In, nystatin, natamitsiny activity of Clotrimazolum can decrease.

Contraindications:

Hypersensitivity to Clotrimazolum or excipients, the I trimester of pregnancy, the menstrual period (for this dosage form).

With CARE lactation Period.

USE DURING PREGNANCY AND DURING FEEDING BY THE BREAST
At clinical and pilot studies it was not established that use of drug during pregnancy or during feeding by a breast exerts negative impact on health of the woman or a fruit (child). However the issue of expediency of purpose of drug has to be resolved individually after consultation of the doctor.

Overdose:

Use of drug in the raised doses does not cause any reactions and states, life-threatening. In case of unforeseen use of drug the following symptoms are (inside) possible: anorexia, nausea, vomiting, gastralgias, abnormal liver function; seldom — drowsiness, hallucinations, a pollakiuria, skin allergic reactions.
There is no specific antidote. It is necessary to appoint inside absorbent carbon. If necessary symptomatic treatment.

Storage conditions:

To store in the dry, protected from light place at a temperature not above 25 °C. To store in the place, unavailable to children!

Tablets vaginal 100 mg. On 3 or 6 tablets place in a blister strip packaging from a film of the polyvinyl chloride and printing aluminum foil varnished. On 6 or 12 tablets place in a container polymeric for medicines. One container or 1, 2, 3, 4, 5 blister strip packagings together with the application instruction place in a pack from a cardboard.
